单相与双相抑郁障碍患者外周血GSK-3β差异表达的研究

目的：探讨单相和双相抑郁患者外周血单核细胞内GSK-3β蛋白的表达差异。材料与方法：从大连市第七人民医院门诊及病房患者中选取双相抑郁障碍患者30例，单相抑郁障碍患者30例。正常对照30例。在受试者知情并同意的前提下，采集其一般信息后，取受试者静脉血20ml并提取其外周血单核细胞。利用免疫印迹法(Wsetern Blotting)检测受试者外周血单核细胞中总GSK-3β及p-GSK-3β蛋白含量。结果：1.单相与双相抑郁障碍组患者的p-GSK-3β/总GSK-3β的比值均较正常组增加，差异有统计学意义(P=0.000)；双相抑郁障碍组患者较单相抑郁障碍组的p-GSK-3β/总GSK-3β的比值增加，差异有统计学意义(P=0.000)。2.单相抑郁障碍患者的p-GSK-3β/总GSK-3β的比值与其HAMD分数两者相关性不存在统计学意义(R=0.325,P=0.08)，而双相抑郁障碍患者的p-GSK-3β/总GSK-3β的比值与其HAMD分数呈正相关(R=0.424,P=0.019)。结论：p-GSK-3β/总GSK-3β的比值变化可能与单相及双相抑郁障碍的病理机制有关，能在早期鉴别单相和双相抑郁障碍发挥重要的作用。据此可以为未来研发治疗单相和双相抑郁障碍的靶向药物提供依据。

| Abstract |
AbstractObjective:To investigate there is differential expression of glycogen synthase kinase-3beta in peripheral blood between unipolar and bipolar depression. Materials and methods:the outpatient and ward patients of Dalian Seventh People’s Hospital Affiliated to Dalian Medical university were selected,including 30 patients of bipolar depression and 30 patients of unipolar depression. And there were 30 cases in the normal group. All participant general states were collected under informed consent, recorded their essential information. And their 20ml peripheral venous blood was collected. The proteins of total GSK-3β and p-GSK-3β in peripheral blood mononuclear cells were measure by Western blotting.Result：1.The ratio of p-GSK-3β to total GSK-3β between the unipolar depression group and bipolar depression group are higher than the normal group, and there was a significant difference (P =0.000< 0.01). The ratio of p-GSK-3β to total GSK-3β in the bipolar depression group was higher than the unipolar depression group, and there was significant difference (P=0.000 < 0.01). 2.The ratio of p-GSK-3β to total GSK-3β in the unipolar depression group has no significant correlation with the their scores of HAMD (r = 0.325, P = 0.08 > 0.05). But the ratio of p-GSK-3β to total GSK-3β in the bipolar depression group was positively correlated with the their scores of HAMD (r = 0.424, P = 0.019 < 0.05).Conclusion：1.The changes of the ratio of p-GSK-3β to total GSK-3β may play a key role in the differntial disagnosis between unipolar and bipolar depression and may involve in the pathogenesis of unipolar and bipolar depression , which may provide a basis for the development of their targeted drugs .
