[Abstract] Objective To investigate the efficacy of agomelatine in the treatment of depression and its effect on cardio pulmonary coupling (CPC) sleep parameters. Methods A total of 100 cases of depression from January 2021-August 2023 were randomized according to the numerical table method, 48 cases in the control group applied sertraline and 52 cases in the observation group applied agomelatine. Mood scores, CPC sleep parameters, and treatment emergent symptom scale (TESS) were compared. Results Hamilton depression scale-17 (HAMD-17) and Hamilton anxiety scale (HAMA) were significantly reduced in both groups before and after treatment (P<0.05). The difference between the groups after treatment was not significant (P>0.05). Total sleep, time asleep, sleep quality scores increased, and falling asleep, light sleep, and hypoventilation index decreased in both groups compared to before and after treatment (P<0.05). And all parameters of the observation group were better than the control group after treatment (P<0.05). The difference in TESS scores between the two groups at 2, 4 and 8 weeks of treatment was not significant (P>0.05). Conclusion Agomelatine for depression and sertraline achieved consistent clinical efficacy and comparable safety, but the former was effective in improving CPC sleep parameters and enhancing sleep quality.
