[Abstract] Objective To explore the influence of electroacupuncture combined with exercise training on IGF-1 and BDNF expressions of rat hippocampus with depression model. Methods 32 rats with SD were divided into the blank group, the model group, the electroacupuncture group, and the electroacupuncture combined with exercise training group, 8 ones in each group. The blank group were raised normally, and the other groups were bulit SD rat depression model by using chronic stress method. The model group were raised normally after modeling, the exercise training group were given exercise training under external stimulus, once a day, all continued for 21d, and the electroacupuncture combined with exercise training group were given electroacupuncture intervention on the basis of exercise training, once a day, acupuncture on Baihui and Taichong for 15min, all continued for 21d. Behavioral changes in rats were observed before modeling and 7d, 21d and after modeling, IGF-1, BDNF, mRNA and protein expressions of rat hippocampus with depression model were detected by using RT-PCR method and Western Blot method. Results At 7d, 21d and after modeling, the horizontal and vertical movements of rats in each group were significantly lower than those before stress (P < 0.05). At 21d after modeling, the horizontal movement and vertical movement in the electroacupuncture combined with exercise training group were significantly higher than those in the model group (P < 0.05), and that in the electroacupuncture combined with exercise training group were significantly higher than those in the electroacupuncture group (P < 0.05), and there were significantly differences when compared with those in the blank group (P < 0.05). The expression levels of BDNF, mRNA and protein in hippocampus of model group were significantly lower than those in the blank group (P < 0.05). The expression levels of IGF-1, mRNA and protein of rat hippocampus in the model group were significantly higher than those in the blank group (P < 0.05). The expression levels of BDNF, mRNA and protein of rat hippocampus in the exercise training group and the electroacupuncture combined with exercise training group were significantly higher than those in the model group (P < 0.05), and IGF-1, mRNA and protein expression levels were significantly lower than those in the model group (P < 0.05), and the indexes were especially obvious in the electroacupuncture combined exercise training group (P < 0.05). Conclusion It has a better therapeutic effect on depression rats of electroacupuncture combined with exercise training, and the mechanism may be related to up-regulation of BDNF and down-regulation of IGF-1 expressions in hippocampus.
