摘要
Complexins蛋白在神经递质释放这一过程中起着关键的调控作用。既往研究显示精神分裂症患者脑部CPLX mRNA及Complexins蛋白水平显著下降。本研究旨在进一步探索CPLX2基因启动子区甲基化与精神分裂症的相关性。方法 采用Taqman探针qPCR (Methylight)技术测定116名男性精神分裂症患者及114名男性健康对照者CPLX2基因启动子区甲基化状况。对精神分裂症患者采用利培酮2~6mg治疗,待患者精神病性症状消失后再次测定CPLX2基因启动子区甲基化状况。结果 精神分裂症患者CPLX2基因启动子区甲基化率显著高于正常对照组(P<0.001);经治疗,精神病性症状消失后,CPLX2基因启动子区甲基化率显著下降(P<0.001)。结论 CPLX2基因启动子区甲基化程度增高与精神分裂症密切相关。
Abstract
Objective Complexins play a critical role in the control of fast synchronous neurotransmitter release. Previous studies have reported a reduction in Complexins mRNA and protein levels in the brains from schizophrenic patients. The aim of this study was to investigate the association of CPLX2 gene promoter methylation with schizophrenia. Methods Taqman probe qPCR (Methylight) technology was used to determine the methylation status of the CPLX2 gene promoter region in 116 male schizophrenia patients and 114 male healthy controls. Patients with schizophrenia were treated with risperidone 2-6 mg, and the methylation status psychotic symptoms. Results The methylation rate of CPLX2 gene promoter region in patients with schizophrenia was significantly higher than that of normal control group, P value <0.001; after treatment, with the remission of psychotic symptoms, the methylation rate of CPLX2 gene promoter region decreased significantly, P<0.001 . Conclusion The increased methylation levels of CPLX2 gene promoter region is closely related with the psychotic state of schizophrenia.
