慢性粒细胞白血病是一类造血干细胞恶性克隆性疾病,其发生机制与组蛋白修饰模式异常相关。本文系统地分析了基因body区域内组蛋白修饰H3K79me2结合丰度与基因差异表达的关系。结果发现,在已有的调控机制外,正常细胞中基因body区域内H3K79me2结合丰度与基因差异表达程度呈负相关(ρ=-0.81,P<0.01)。即:在正常细胞内,H3K79me2结合丰度较低的基因,它们在癌变过程中表达水平差异程度较大;反之,表达水平差异程度较小。调控元件分析显示,转录因子GATA3、GATA4、TEAD1、TEAD3、TEAD4和TRPS1促进了富含低H3K79me2结合丰度基因表达的上调(ρ=0.95, P<0.01);而转录因子Ascl2、E2A、IRF3、IRF4、Oct4和ZEB2则诱导了富含低H3K79me2结合丰度基因的表达下调(ρ=0.97, P<0.01)。进一步的分析揭示,H3K79me2结合丰度对基因差异表达的负调控作用不受基因表达水平和基因长度的影响。通路富集分析表明,富含低H3K79me2结合丰度的基因参与了白细胞迁移调节、细胞间粘附等与白血病相关的信号通路。这些发现共同暗示,基因body区域内H3K79me2的低结合丰度可能与慢性粒细胞白血病的发生密切相关。
Abstract
Chronic myelogenous leukemia (CML) is a malignant clonal disease of hematopoietic stem cells, and studies have shown that the progression of CML is associated with abnormal histone modification (HM) patterns. Here, we explored the relationship between H3K79me2 intensities in the gene body region and gene expression changes. The results exhibited that the binding intensities of H3K79me2 in the normal cells were negatively correlated with gene expression changes beyond the regulatory mechanisms that have been discovered (ρ=-0.81, P<0.01). Specifically, the expression levels of gene with low H3K79me2 binding intensities changed significantly, while the expression levels of gene with high H3K79me2 binding intensities changed little during carcinogenesis. Regulatory element analysis displayed that transcription factors GATA3, GATA4, TEAD1, TEAD3, TEAD4 and TRPS1 promoted the up-regulation of genes with low H3K79me2 intensities (ρ=0.95, P<0.01), and transcription factors Ascl2, E2A, IRF3, IRF4, Oct4 and ZEB2 induced the down-regulation of genes with low H3K79me2 binding abundance (ρ=0.97, P<0.01). Further analysis revealed that H3K79me2 intensities negatively regulated gene expression changes regardless of gene expression levels and gene lengths. Finally, Pathway enrichment analyses showed that genes with low H3K79me2 intensities were involved in leukemia-related signaling pathways, such as regulation of leukocyte migration, regulation of cell adhesion. These conclusions suggested that lower gene-body H3K79me2 intensities may be closely related to the occurrence of CML.
