摘要
【目的】本试验旨在探究小麦白粉病菌(Blumeriagraminisf.sp.tritici)侵染对小麦幼苗生长与生理指标的影响,揭示不同抗性小麦材料响应白粉病菌侵染的生理防御机制。【方法】以小麦抗病品系7D49和感病品系薛早为试验材料,于二叶一心期接种白粉病菌生理小种E09,测定接菌前(0d)及接菌后第2、4、6、8、10d幼苗的苗长和茎粗,叶片的叶绿素含量,超氧化物歧化酶(SOD)、苯丙氨酸解氨酶(PAL)、过氧化氢酶(CAT)、过氧化物酶(POD)、多酚氧化酶(PPO)的活性及丙二醛(MDA)含量。【结果】接菌后各时间点,7D49的苗长显著高于薛早(P<0.05),茎粗比薛早粗;随着接菌时间的延长,7D49和薛早叶片的叶绿素含量均显著降低,且同一时间点,7D49的叶绿素含量显著高于薛早(P<0.05);7D49的MDA含量与接菌前没有显著性差异,而薛早的MDA含量则显著降低(P<0.05)。防御酶活性分析表明,接菌后,7D49的SOD活性显著升高,第4d达到峰值,随后逐渐降低,而薛早的SOD活性则显著下降(P<0.05);7D49和薛早的CAT和PAL活性均显著降低(P<0.05),PPO活性均先降低随后上升,POD活性无明显变化规律。【结论】抗病品系7D49生长受白粉病菌侵染影响较小,接菌后,苗长和叶片叶绿素含量和SOD活性均显著升高,从而抵御白粉病菌侵染。该研究结果为探讨小麦白粉病菌与小麦的互作机理提供了一定理论参考依据。
Abstract
【Objective】This study aims to investigate the effects of wheat powdery mildew (Blumeria graminis f. sp. tritici) infection on the growth and physiological parameters of wheat seedlings, elucida- ting the physiological defense mechanisms of wheat varieties exhibiting varying levels of resistance to this pathogen. 【Method】The experiment utilized a disease-resistant line (7D49) and a susceptible line (Xu- ezao). At the two-leaf-one-heart stage, seedlings were inoculated with B. graminis f. sp. tritici race E09. Measurements were recorded prior to inoculation (0 day) and at intervals of 2, 4, 6, 8, and 10 days post-inoculation (dpi). Key parameters included seedling shoot length, stem diameter, leaf chloro- phyll content, enzymatic activities of superoxide dismutase ( SOD ), phenylalanine ammonia-lyase (PAL), catalase (CAT), peroxidase (POD), and polyphenol oxidase (PPO), as well as malondialde- hyde (MDA) content. 【Result】After inoculation, the shoot length of 7D49 was significantly greater than that of Xuezao, accompanied by a thicker stem diameter. Although chlorophyll content decreased signifi- cantly in both lines over time, 7D49 consistently exhibited higher chlorophyll levels compared to Xuezao at each measured time point (P<0. 05). Notably, the MDA content in 7D49 remained stable compared to pre-inoculation levels, while Xuezao displayed a significant decline (P<0. 05). Analysis of defense enzyme activities indicated that SOD activity in 7D49 significantly increased, peaking at 4 dpi before gradually declining, whereas Xuezao experienced a marked reduction in SOD activity (P<0. 05). Addi- tionally, significant decreases were observed in both CAT and PAL activities across both lines ( P < 0. 05). PPO activity initially decreased before rising in both varieties, while POD activity displayed no discernible trend. 【Conclusion】The resistant line 7D49 demonstrated a reduced impact from powdery mildew infection on growth parameters. After inoculation, 7D49 maintained significantly a notable in- crease in SOD activity, contributing to its resistance against B. graminis f. sp. tritici. These findings provide critical insights into the interactions between wheat and the powdery mildew pathogen.
