摘要
【目的】通过建立大花绣球‘蓝色妈妈’的高效稳定快繁体系,以提高其繁殖效率。【方法】以大花绣球‘蓝色妈妈’带腋芽茎段为外植体,探究不同浓度植物生长调节剂对芽的诱导、丛生芽增殖和生根的影响,以及不同移栽基质对移栽成活率的影响。【结果】(1)诱导培养阶段,随着6-BA和NAA浓度升高,诱导率呈下降趋势,当6-BA为0.5mg/L、NAA为0.1mg/L时,诱导率最高(47.8%),最佳诱导培养基为MS+0.5mg/L6-BA+0.1mg/LNAA。(2)增殖培养阶段,增殖率与6-BA的浓度成正比,最佳增殖培养基为MS+2.0mg/L6-BA+0.2mg/LNAA,增殖系数最高为3.16。(3)生根培养阶段,随IBA浓度的升高,生长状况呈先上升后下降的趋势,当IBA浓度为0.4mg/L时,生根率达100%(平均根长为4.07cm,主根数为12.3);(4)在蛭石∶珍珠岩的体积比为1∶1的移栽基质中,成活率达100%,植株长势强,株高和冠幅均高于其他处理。【结论】建立了大花绣球’蓝色妈妈’高效离体快繁体系,为其规模化生产提供了技术基础。
Abstract
【Objective】It is to improve the reproductive efficiency of Hydrangea macrophylla ‘Blue Mama’ by establishing an efficient, stable and rapid propagation system. 【Method】The effects of different concentrations of plant growth regulators on the germination, proliferation, and rooting of axillary buds and the effect of different substrate ratios on seedling transplantation were investigated using H. macrophylla ‘Blue Mama’ with axillary bud stem segments as explants. 【Result】(1) In the induction culture stage, the induction rate showed a downward trend as the concentrations of 6-BA and NAA in- creased. When 6-BA was 0. 5 mg / L and NAA was 0. 1 mg / L, the highest induction rate (47. 8% ) was obtained. The optimal induction culture medium was MS+0. 5 mg / L 6-BA+0. 1 mg / L NAA. (2) In the proliferation culture stage, the most suitable proliferation medium was MS+2. 0 mg / L 6-BA+0. 2 mg / L NAA, the proliferation rate was positively proportional to the concentration of 6-BA, and the highest pro- liferation coefficient was 3. 16. (3) During the rooting culture stage, with the increase of IBA concentra- tion, the growth condition exhibited a trend of initially ascending and then descending. When the IBA concentration was 0. 4 mg / L, the rooting rate reached 100% (root length was 4. 07 cm and number of main roots was 12. 3). (4) In the transplanting substrate with a volume ratio of 1 ∶1 of vermiculite to perlite, the survival rate was 100% , the plant growth was vigorous, and both the plant height and crown width were higher than those in other treatments. 【Conclusion】A high efficient propagation system for H. macrophylla ‘Blue Mama’ has been established, which provides some theoretical references for the rapid propagation of Hydrangea macrophylla.
