朱顶红‘糖果仙女’组培快繁技术研究

摘要
【目的】通过对朱顶红‘糖果仙女’的鳞茎进行组培快繁技术优化研究,初步建立朱顶红‘糖果仙女’无菌培养体系,为朱顶红优质种球的高效培育提供理论依据,为进一步提升朱顶红的产业化生产水平奠定基础。【方法】以朱顶红‘糖果仙女’鳞茎为材料,采用不同的消毒时间、不同培养基配方等,探讨不同因素对朱顶红‘糖果仙女’外植体消毒、芽诱导、增殖及生根等的影响。【结果】不同的消毒时长对朱顶红‘糖果仙女’鳞茎的消毒效果存在差异,当以75%酒精消毒时间60s,2%NaClO溶液消毒12min时,鳞茎的存活率最高,为73.3%;在不同培养基的激素配比中,鳞茎芽诱导培养基以MS+1.0mg/L6-BA+1.5mg/LNAA+2.0mg/LTDZ+1g/LAC的诱导效果最好,诱导率达到75.32%;增殖培养基以MS+2.0mg/L6-BA+1.5mg/LNAA+1.0mg/LTDZ+1g/LAC的增殖效果最好,增殖系数为5.28;生根培养基以MS+0.5mg/LIBA+1.0mg/LNAA+1g/LAC的生根效果最好,平均生根数5.6条,平均生根长度6.4cm,生根率达到了96.42%。【结论】朱顶红‘糖果仙女’无菌培养体系最适宜消毒方式为75%酒精消毒60s+2%NaClO溶液消毒12min;最适宜鳞片小鳞茎芽诱导培养基为MS+1.0mg/L6-BA+1.5mg/LNAA+2.0mg/LTDZ+1g/LAC;增殖培养基为MS+2.0mg/L6-BA+1.5mg/LNAA+1.0mg/LTDZ+1g/LAC;生根培养基为MS+0.5mg/LIBA+1.0mg/LNAA+1g/LAC。
Abstract
【Objective】Through the study on the tissue culture and rapid propagation of the bulbs of Hippeastrum vittatum ‘Candy Nymph’, the aseptic culture system was established, to provide a theoreti- cal basis for the efficient breeding of high-quality bulbs of H. vittatum, and lay a foundation for further promoting the industrialization production of H. vittatum. 【Method】The bulbs of H. vittatum ‘ Candy Nymph’ were used as materials, and different disinfection time and culture media were used, to investi- gate the effects of different factors on disinfection, shoot induction, proliferation and rooting of H. vitta- tum ‘Candy Nymph’. 【Result】The disinfecting effects of different disinfecting time on the bulbs of H. vittatum ‘ Candy Nymph’ were different. When the disinfecting time was 60 s with 75% alcohol and 12 min with 2% NaClo, the survival rate of the bulbs was the highest, being 73. 3% ;Among the different media, the induction rate of bulb bud induction medium with MS+1. 0 mg / L 6-BA+1. 5 mg / L NAA+ 2. 0 mg / L TDZ+1 g / L AC was the best, reaching 75. 32% ;MS +2. 0 mg / L 6-BA+1. 5 mg / L NAA+ 1. 0 mg / L TDZ+1 g / L AC was the best medium for proliferation, and the proliferation coefficient was 5. 28; The best rooting medium was MS+0. 5 mg / L IBA+1. 0 mg / L NAA+1 g / L AC, with an average rooting number of 5. 6, an average rooting length of 6. 4 cm and a rooting rate of 96. 42% . 【Conclusion】 The most suitable disinfection method for the sterile culture system of H. vittatum ‘Candy Nymph’ was 75% alcohol disinfection for 60 seconds+2% NaClO solution disinfection for 12 minutes; The optimum medium for bulblet induction was MS+1. 0 mg / L 6-BA+1. 5 mg / L NAA+2. 0 mg / L TDZ+1 g / L AC. The proliferation medium was MS+2. 0 mg / L 6-BA+1. 5 mg / L NAA+1. 0 mg / L TDZ+1 g / L AC, and the rooting medium was MS+0. 5 mg / L IBA+1. 0 mg / L NAA+1 g / L AC.
