药茶用菊花‘婺源皇菊’组培快繁体系研究

摘要
【目的】本研究旨在建立‘婺源皇菊’的组培快繁体系,为规模化生产药茶用菊花‘婺源皇菊’种苗提供技术支撑。【方法】以‘婺源皇菊’幼嫩茎段为外植体,次氯酸钠为消毒剂,消毒时间分别为5(A1)、10(A2)、15(A3)和20(A4)min,以MS培养基为基本培养基,并添加不同浓度组合的植物生长调节剂,对外植体的消毒、初代培养、继代培养、生根培养、炼苗移栽过程进行了研究。【结果】不同消毒时间、不同植物生长调节剂配比对‘婺源皇菊’外植体初代培养效果影响显著。不同消毒时间处理对外植体褐化率和染菌率的影响分别为:A1A2>A3>A4;初代培养时添加6-BA2.0mg/L+NAA0.2mg/L+KT0.1mg/L的培养基芽诱导率、出芽指数及苗高均为最高;继代增殖培养时则为添加6-BA1.5mg/L+NAA0.2mg/L+KT0.1mg/L的培养基芽诱导率和出芽指数最高;生根培养时添加6-BA0.5mg/L+NAA0.3mg/L+IBA0.2mg/L的组培苗生根率最高、单株平均根数最多、单株平均根长最长;最适宜的炼苗天数为6~7d。【结论】外植体最佳消毒方式为70%酒精消毒10s后,用2%次氯酸钠消毒10min;以MS为基础培养基,初代培养最佳植物生长调节剂组合为6-BA2.0mg/L+NAA0.2mg/L+KT0.1mg/L,继代培养最佳组合为6-BA1.5mg/L+NAA0.2mg/L+KT0.1mg/L;生根培养的基本培养基为1/2MS,最佳植物生长调节剂组合为6-BA0.5mg/L+NAA0.3mg/L+IBA0.2mg/L。本研究初步构建了‘婺源皇菊’的组培快繁技术流程,为‘婺源皇菊’种苗的规模化繁殖提供了理论基础与技术支撑。
Abstract
【Objective】The aim of this study is to establish a fast breeding system of ‘ Wuyuan- huangju’, and to provide technical support for the large-scale production of ‘ Wuyuanhuangju ’ seedlings. 【Method】The disinfection, primary culture, secondary culture, rooting culture and seedling transplantation process of ‘Wuyuanhuangju’ were studied by using young stem segments of ‘Wuyuan- huangju’ as the explant, hypochloric acid as the disinfectant, the disinfection times were 5 (A1), 10 (A2), 15 (A3), and 20 (A4) minutes, respectively, and different plant growth regulator combinations added with MS culture medium as the basic culture medium. 【Result】Different sterilization time and dif- ferent plant growth regulator ratios had significant effects on the primary culture effect of ‘ Wuyuan- huangju’ explants. The ratios of browning and bacterial contamination of tissue culture seedling in differ- ent treatments caused by sterilization time were: A1A2>A3>A4 respectively; The me- dium supplemented with 6-BA 2. 0 mg / L+NAA 0. 2 mg / L, and KT 0. 1 mg / L had the highest bud induc- tion rate, germination index, and seeding height during the initial cultivation; The medium supplemented with 6-BA 1. 5 mg / L, NAA 0. 2 mg / L, and KT 0. 1 mg / L showed the highest bud induction rate and germination index during subculture; The tissue culture seedlings with added 6-BA 0. 5 mg / L + NAA 0. 3 mg / L+IBA 0. 2 mg / L had the highest rooting rate, the highest average number of roots per plant, and the longest average root length per plant during rooting cultivation; the optimal number of days of seedling refining was 6 ~ 7 days. 【Conclusion】The optimal disinfection method was 70% alcohol disin- fection for 10 seconds, followed by disinfection with 2% hypochloric acid for 10 minutes; the optimal pri- mary culture medium MS+6-BA 2. 0 mg / L+NAA 0. 2 mg / L+KT 0. 1 mg / L; optimal successional medium MS+6-BA 1. 5 mg / L+NAA 0. 2 mg / L+KT 0. 1 mg / L; optimal rooting medium 1 / 2MS+6-BA 0. 5 mg / L+ NAA 0. 3 mg / L+IBA 0. 2 mg / L. This study preliminarily constructed the tissue culture and rapid propa- gation technology process of ‘Wuyuanhuangju’, providing theoretical basis and technical support for the large-scale propagation of ‘Wuyuanhuangju’ seedlings.
