摘要
【目的】为了扩大龙牙百合בBlackStone’杂种株系LB-2的群体数量,建立及优化龙牙百合杂种后代离体再生体系。【方法】本研究通过正交试验设计,探讨了不同激素种类及浓度对百合鳞片不定芽诱导和鳞茎膨大生根的影响。【结果】百合鳞片在配方为MS+1.0mg/LNAA+1.5mg/L6-BA+1.0mg/LKT+30g/L蔗糖+7.5g/L琼脂培养基中的平均出芽数和不定芽诱导率最高,分别为2.33个和93.33%;在鳞茎膨大倍数和根系生长状况方面,百合鳞茎在配方为MS+0.2mg/LNAA+0.5g/LAC+80g/L蔗糖+7.5g/L琼脂培养基中的膨大倍数和生根效果最优,鳞茎膨大倍数为2.4,并且根系粗壮且较多。【结论】本研究通过激素浓度组合及优化激素配置,优化了从鳞片诱导成芽到小鳞茎生长膨大生根整个过程的组培离体再生体系,该体系可为今后龙牙百合杂交后代的组培繁育和推广应用提供技术支持。
Abstract
【Objective】It is to expand the population of the hybrid strain LB-2 of Lilium brownii var. viridulum Baker× ‘ Black Stone’ and establish and optimize the in vitro regeneration system of Lilium brownii var. viridulum Baker hybrid offspring. 【Method】This study investigated the effects of different hormone types and concentrations on the induction of adventitious buds and root enlargement of lily scales through orthogonal experimental design. 【Result】The average number of buds and the induction rate of adventitious buds of lily scales were highest in the medium formula of MS medium supplemented with 1. 0 mg / L NAA, 1. 5 mg / L 6-BA, 1. 0 mg / L KT, 30 g / L sucrose, 7. 5 g / L agar, with values of 2. 33 buds and 93. 33% , respectively; from the swelling ratio of the bulbs and the growth status of the root sys- tem, it was found that the lily bulbs had the best swelling ratio of the bulbs and rooting effects in the me- dium formula of MS medium supplemented with 0. 2 mg / L NAA, 0. 5 g / L AC, 80 g / L sucrose, 7. 5 g / L agar. The bulb swelling ratio was 2. 4, and the root system was thick and abundant. 【Conclusion】This study optimized a tissue culture in vitro regeneration system for the entire process from scale induction into buds to small bulb growth, enlargement, and rooting through hormone concentration combinations and optimized hormone configurations. This in vitro regeneration system can provide technical support for the future tissue culture breeding and promotion application of hybrid offspring of Lilium brownii var. viri- dulum Baker.
