[Abstract] Objective: To explore the improvement effect of Duloxetine and Sertraline on depression in elderly patients with depression. Methods: The case data from October 2021 to October 2022 were collected for research. The selected subjects were elderly patients with depression who were admitted to the hospital. A total of 100 patients were included. According to the different treatment methods, they were divided into the control group (treated with Sertraline) and the observation group (treated with Duloxetine), and 50 patients were included in each group; Observe and compare the degree of depression, sleep quality, clinical efficacy, and serum factors between the two groups. Result: After 1 week, 4 weeks, and 6 weeks of treatment, the Hamilton depression rating scale（HAMD） score of the observation group was significantly lower than that of the control group (P<0.05); After treatment, the Pittsburgh Sleep Quality Index（PSQI） scores of both groups were lower than before treatment, and the observation group was lower than the control group (P<0.05); The total clinical effective rate of the observation group was higher than that of the control group (P<0.05); After treatment, the levels of norepinephrine（NE）, 5-hydroxytryptamine（5-HT）, anddopamine（ DA） in both groups showed varying degrees of increase, and the observation group showed a greater increase than the control group (P<0.05). Conclusion: Compared with Sertraline, Duloxetine can achieve more significant therapeutic effects in elderly patients with depression, and improve the degree of depression and Sleep disorder.
