Objective: The purpose of this study was to investigate the effects of Olanzapine combined with Fluvoxamine on the dosage of Olanzapine, N-desmethyl Olanzapine, and serum prolactin levels in schizophrenia patients with depression. Methods:The study selected 96 patients diagnosed with schizophrenia with depression admitted to the hospital from January 2021 to November 2022 as the research subjects. They were randomly divided into two groups: Olanzapine treatment group (control group, n=48) and Olanzapine combined with Fluvoxamine treatment group (observation group, n=48). The clinical efficacy, Olanzapine dosage, N-desmethyl Olanzapine concentration, serum prolactin levels, and treatment safety were compared between the two groups. Results: The total effective rate of the experimental group was higher than that of the control group after treatment (p<0.05). The negative and general psychopathology scores on the Pasitive and negative syndrome scale（PANSS）in the experimental group were lower than those in the control group (p<0.05). The Hamilton Depression Scale-17 (HAMD-17) score in the experimental group was also lower than that in the control group (p<0.05). The Olanzapine dosage, N-desmethyl Olanzapine concentration, and serum prolactin levels in the experimental group were lower than those in the control group (p<0.05). The incidence of adverse reactions in both groups was similar (p>0.05). Conclusion: Combined treatment with Olanzapine and Fluvoxamine can improve the negative symptoms and depressive symptoms in schizophrenia patients with depression. The dosage of olanzapine is lower, reducing N-demethylolanzapine and serum prolactin levels The treatment is safe and effective.
