论文观察氨氯地平预处理对氧化型低密度脂蛋白诱导的平滑肌细胞泡沫化过程中亲环素A表达的影响及其对胆固醇蓄积的作用,从新的角度探讨氨氯地平的抗动脉粥样硬化作用。结果氧化型低密度脂蛋白处理组的细胞内亲环素A的表达明显减少;经氨氯地平处理后,随着氨氯地平浓度的增加,细胞内亲环素A的表达逐渐增加,10.0μmol/L氨氯地平预处理细胞组效果最明显,较氧化型低密度脂蛋白组差异有显著性(P<0.05)。油红O染色显示,氧化型低密度脂蛋白组细胞内大量脂滴形成,细胞内胆固醇酯/总胆固醇的比值为57.9%,符合泡沫细胞特征;预先予氨氯地平处理后,随着药物浓度的增加,细胞内脂滴逐渐减少,细胞内胆固醇酯/总胆固醇的比值逐渐降低,10.0μmol/L氨氯地平预处理细胞组效果最明显,为37.8%;结论氨氯地平预处理,可上调氧化型低密度脂蛋白诱导的平滑肌细胞泡沫化过程中亲环素A的表达,减轻细胞内的胆固醇蓄积。

| Abstract |
The study investigated the effects of amlodipine pretreatment on the expression of Cyclophilin A during the foam cell formation of smooth muscle cells induced by oxidized low-density lipoprotein (ox-LDL) and its impact on cholesterol accumulation, providing a novel perspective on the anti-atherosclerotic mechanism of amlodipine. The results showed that the expression of intracellular Cyclophilin A was significantly reduced in the ox-LDL-treated group. Following amlodipine treatment, the expression of intracellular Cyclophilin A gradually increased with rising amlodipine concentrations, with the 10.0 μmol/L amlodipine pretreatment group showing the most pronounced effect, exhibiting a statistically significant difference compared to the ox-LDL group (P < 0.05). Oil Red O staining revealed that the ox-LDL group exhibited extensive lipid droplet formation within the cells, with an intracellular cholesteryl ester to total cholesterol ratio of 57.9%, consistent with foam cell characteristics. With increasing concentrations of amlodipine pretreatment, intracellular lipid droplets gradually decreased, and the ratio of intracellular cholesteryl ester to total cholesterol progressively declined, with the most significant effect observed in the 10.0 μmol/L amlodipine pretreatment group, reaching 37.8%. In conclusion, amlodipine pretreatment upregulates the expression of Cyclophilin A during ox-LDL-induced foam cell formation in smooth muscle cells and alleviates intracellular cholesterol accumulation.
